Twelvemile Lake is located on S bank of Yukon River, 0.5 mi. S of Twelvemile I. and 9 mi. ENE of Tanana, Yukon-Tanana High.

Local name reported in 1954 by R. E. Isto, U.S. Geological Survey (USGS); so named because of its relationship to Twelvemile I.
